Culturing Journal DataSheet
This first post should be updated regularly to include new information as events take place or changes are made to your system
General Species: Nannochloropsis sp.
Species description: Non Motile golden brown algae, spherical cell average 5 micron diameter ranges from freshwater to marine (lower salinities)
Culture source (link if possible): www.florida-aqua-farms.com If algae, CCMP # (ref http://ccmp.bigelow.edu/ ): CCMP531
https://ccmp.bigelow.org/node/1/strain/CCMP531 Culturing Vessel Details Salinity: 1.019
Temperature: Ambient 72*
Vessel description: Cylindrical 3 liter arrowhead water jugs, dimple in bottom
Lighting description: three 4 foot dual tube normal output flourescents mounted vertically on wall behind culture shelves (approx 4 inches from vessels) Sylvania 3500K bulbs currently used
Lighting cycle: 14 hours light 10 hours dark
Aeration description: Aeration provided by rigid airline sunk to the bottom of each culture rack, no airstone is used, bubble rate is set semi rapid to provide for water to circulate at approximately 1 rotation per second
Methodologies Split methodology: Each culture is harvested at split time, culture is then re-started using 40% of original volume with harvested 60% being replaced with ASW mixed to 1.019 and 5 ml of Guillards F formulation (provided by florida aqua farms) is added. Splits are performed every 8 days.
Culture medium description: 5ml Guillards F formulation (Micro Algae Grow from Florida Aqua Farms)
Cell count: average 22 million cells per ml via hemocytometer cell count method, 1.5cm on seechi stick
Reference links: Additional Information Notes: Best beginner species and a staple in aquaculture, while nutritional profiles and cell sizes are not exactly perfect it is a good fall back and rapid producer. Culture strains have been maintained for more than one year now. Number of cultures maintained totals 9 (8 standards and one mother) Mother culture is split on a 18 day schedule receiving full F strenght (6ml per 3 liters)
