Methodologies
Split methodology: As culture matures (8-10 days at my current room temp), 1/3 of bottle used to start new culture, 1/3 used as food (presently for my coral frag tank), and then the bottle topped off with new salt water (1.020 SG).  Cultures fertilized with Guillard's at standard f/2 dosing (per bottle instructions).

Culture medium description:
New saltwater (Instant Ocean or Reef Crystals mix) at 1.020 SG, made with tap water.  No additional bleach added for sterilization.  Dechlorination is done with AmQuel just prior to use, with Guillard's added at that time as well (10 drops per liter).

Cell count:
 not measured at this time

Ok, I've taken the plunge to start culturing phyto.  I am hydrating my culture disk today, will place into the culture bottle tomorrow.  Hopefully this time next week I have a nice green bottle to split.

Good luck, hope that you will have better success than my only partial success at starting a culture.

The first culture split today.  The top photo was of the initial culture start-up from the FAF disk.  The bottom is the new 2 liter culture.  I'm going to just keep a few bottles running at present, since I'm not feeding any rotifers right now.

Cultures continue to grow well. I'm currently keeping 4 bottles active at a time, starting a new bottle about every 3 days. My frag tank gets the harvest at that time.

Day 94 as I count them. Continuing to maintain 4 bottles, new one started approximately every 2-3 days (using about 1/3 of the oldest bottle). Using 12-15 drops f/2.
