Culturing Journal DataSheet This first post should be updated regularly to include new information as events take place or changes are made to your system General Phyto Species: Nannochloripsis Species description: Very dense Culture source (link if possible ): Original culture was from another hobbyist If algae, CCMP # (Optional ): http://ccmp.bigelow.edu/ Culture Establishment Date: End of Feb. 2013 Continuation Date: (Batch 1) 4/15, 4/29, ?, 5/29 (Batch 2) 4/22, 5/5, 5/26, split dates are no longer consistent and for me at least this is working Culturing Vessel Details Salinity: 1.019 Temperature: room temp pH: unknown Vessel description: 2 1/2 L bottles Lighting description: 6,500 K T-5 Lighting cycle: 6am-10pm Aeration description: Good strong bubble Methodologies Split methodology: It was suggested to me that I should wait a month or 2 to split for denser cultures. They are being split monthly. I take 2 bottles when I split, one of finished product and one to be split between 3 new bottles. I have let bottles run up to a month and a half without issue. Culture medium description: Micro Algae grow10 drops per litter now changed to 5 drops of F/S per 2 littler or 3 mls of miracle grow per the same. Must also include that I re add the fertilizer weekly to the bottles Cell count: (if known) Reference links: his culture came from a hobbyist and he was running them up to 4months before split. Additional Information (No Pictures or Videos in the Section Please) Notes: About 2 months ago I was having 3 out of 5 bottles crash on me, and tried everything from using tap instead of RO, changing the SG and adding a fan foe air flow and better PH. urns out non of this was the problem and as soon as I started adding less fertilizer more often all my crashes stopped. Updated pictures below You will be required to provide photographic evidence and as much detail as possible about your project in this thread. If your thread does not contain detailed enough photos and information the MBI Council will not be able to approve your reports.

Those cultures are very dark! I wouldn't split based on time but rather how dense/dark. I would be concerned that one day too long and your cultures could crash out.

That's what I would assume, these cultures are very dark. I'm going to split a few off I think and see how long it takes one from start to crash.

I just let my Iso get too dark and it crashed. Now I'm having a hard time starting it back up. Is it possible or do I need to get a new culture start?

I tried starting my last one back up after it crashed and didn't have any luck. That was a FAF disk and this is a culture I got from a local. The local culture is much better, and I never got the other one close to as dense. I would see if anyone local has some you could get.

I have had luck sieving the Iso with a 40uM mesh and then restarting...but it was done before it went completely clear. It's worth a shot, but I wouldn't bank on it.

I personally split my cultures on a 7 day cycle. If they have not reached full density within 7 days, then I know the culture is struggling. I have done this 7 day cycle for multiple types of phyto, and have not had any problems.

That's how I was doing it w/ my last try. It was also The FAF disk, so not sure if that was it or not. Cultures were strong in the beginning, but that only lasted 3 months.

So after splitting and giving a culture away last week I was left w/ 3 going. Than my kitties knocked one over and I had to fix their mistake. I'm left w/ 1 culture I started on the 15th and one started today.

What do you mean, top off...like for evaporation? I keep my containers with lids or some other filter/blocking to limit cross contamination, and that limits evaporation. Edit: I just looked at your pics. Are you really getting a lot of evaporation with lids on? How long between splits?

They are lidded, but yes for evaporation as I will be keeping them going for closer to a month and there is some evap over that time.

They are not as dark as I would like and I am trying out some advice from the person I got the culters from. Believe this or not, they have been running theirs for up to 4 months before splitting them and they are some of the best cultures I've seen.

Split today, I think I am going to stick to a 2 week split scheduled, they are very dark at the 2 week mark and I probably could get a few more days out of them. I'll save that experiment for when I have a large supply though. Finished product New cultures

As of right now I have forgotten one of the dates from batch 1 but have since split again. I also did something a little different w/ each batch. Instead of just using the hobby F/S I split batch 2 w/ miraclegrow , and Batch 1 w/ the normal stuff. Dates are 3 days apart here is the result Batch 1 Hard to see but there is a huge difference Batch 2 You can see one of the bottles from the other batch on the right]

Not running them as long as I would like or getting them as dark as I want, but it is a work in progress

Just curious: You seem to equate "dark" with "good". Why? I equate "nutritious" with "good".
