Culturing Journal DataSheet This first post should be updated regularly to include new information as events take place or changes are made to your system General Species: Isochrysis galbana Species description: motile brown algae Culture source (link if possible ): Neptunes Gardens If algae, CCMP # (Optional ): http://ccmp.bigelow.edu/ Culture Establishment Date: 3/12/14, restarted 7/23/14 Continuation Date: 11/1/14 Culturing Vessel Details Salinity: 1.020 Temperature: ambient room temp, approx. 70 degrees F pH: not directly measured, but new mix is generally about 8.0-8.1 Vessel description: 1 quart plastic bottles, with plans to expand into 2 liter plastic bottles as cultures progress Lighting description: 13w spiral cfl bulb Lighting cycle: 16 hours light, 8 hours dark Aeration description: rigid airline tubing with moderate flow Methodologies Split methodology: 2/3 to 3/4 of mature culture used for feeding, remainder to inoculate new culture bottle Culture medium description: initially used both microwave and bleach sterilization techniques. Currently only using bleach sterilization process: 1ml chlorox bleach in 1 gallon NSW, then 1quart culture bottle filled 2/3 to 3/4 full. 20 drops Guillards f/2 and 0.4ml Drs. Foster & Smith Chlorine Neutralizer added per quart bottle. Foster & Smith Chlorine Neutralizer works well for me and is now my sole method for culture medium prep You will be required to provide photographic evidence and as much detail as possible about your project in this thread. Just received my starter culture yesterday and set up 2 bottles. The bottles look darker in the lower picture (with nanno on right) due to camera and lighting angle.

New SW (Reef Crystals or Instant Ocean) prepared at 1.020. My 2 methods of sterilization of the new SW will be as follows: A) One quart bottles (the only ones that fit in my microwave) will be microwaved to temp of 180-190 degrees F. These are then cooled to room temp while covered, and used within 24 hours. B) Bleach sterilization uses standard household bleach at 1 ml/gallon SW prepared usually about 1 week in advance, neutralized with AmQuel immediately prior to use. This has the advantage of allowing larger bottles than I can get into my microwave. I'll see which one of these techniques works best for me. The bleach process is actually easier, but requires additional additives (bleach & AmQuel).

So far the cultures have been doing OK. I have made a few splits. When a bottle gets to a nice tea color, I inoculate new culture medium with about 1/3 mature culture. My initial culture was thus split into 3 more bottles, and I am now up to 4 total bottles running. I have found the "bleach sterilization method" (above) to be easy and effective for me thus far. I did increase the amount of Guillards f/2 to 15 drops per liter, and seem to have better/faster growth.

Unfortunately had a crash. The ISO culture was invaded by nanno or chlorella, and I had to restart about 6 weeks ago. I need to update the journal to reflect dates.

Does the Amquel also neutralize ammonia? You may not want to use that in your cultures. At one time, I was dechlorinating with ChloraM X, and was having trouble, and my thought at the time, was that the Cloram X was messing with the fertilizer. I use Sodium thiosulfate, and have not had problems. I never tested this hypothesis the right way, so it might not be true, but if I was having problems, I might think about it.

Thanks for the suggestion. I may give that a try, as I may be developing the same problem again. If I remember correctly from the label, AmQuel Plus does neutralize ammonia as well at chlorine & chloramines. I'll double check.

Ok, I have restarted the culture on 7/23/14 with a batch brought back from the conference by Waldend. The pic is taken after about 5 days and th fist split. They are really growing nicely. The cultures on the right are tetraselmis, obtained at the same time. I'm using a different dechlor agent (one that doesn't neutralize ammonia). So far I have split the culture 5 times to date.

Since restarting (again!) on 7/23/14, I'm trying a different dechlorination product (Drs. Foster &Smith Chlorine Neutralizer), Which specifically states on the bottle it does not neutralize ammonia, just chlorine and chloramines. I have been using this product with half of my bottles, the other half getting the culture media sterilized by microwaving to 180 degrees F. Thus far, both techniques have been giving me good results. I can't tell any difference in growth rates.

Cultures continue to do great. I stopped using microwave sterilization as the bleach method (& DFS chlorine neutralizer) is working fine. The end of this month will mark the 90 day point since the restart. Here is a pic taken this morning of the 3 bottles I am currently running. The far left was just started yesterday.

Do you still have cultures going strong? Any modifications to your system? I am wanting to start and Iso culture so I am learning as much as I can in order to have a better chance of success.

No changes. I still use the chlorine sterilization method. Cultures are mature and ready to split in 7-9 days.
