Weiss) If algae, CCMP # (Optional ): [link= http://ccmp.bigelow.edu/]http://ccmp.bigelow.edu/[/link] Culture Establishment Date:7/24/2014 Continuation Date: 12/31/2014 Culturing Vessel Details Salinity: 1.024 currently, 1.020 initially. Temperature: Ambient Room Temp (70°F-78°F in summer, 60°F-70°F in winter) pH:Not monitored Vessel description: Current is 2L clear plastic container with screw on lid. Hole drilled in lid to snugly fit a rigid airline. Initial was 1 Quart Wide-mouth canning jars. Plastic wrap cover held with rubberband around perimeter. Rigid airline poked through wrap to feed air and a small puncture for air release. Lighting description: Single T8 bulb under-cabinet lighting fixture. Lighting cycle: 18 on, 6 off. Off is in the afternoon to reduce temp during the warmest part of day (summer and winter) to keep a more consistent temperature. Aeration description: Light boil to keep algae fully in suspension Methodologies Split methodology: Plan is to remove 75% every 7-10 days and add the remaining 25% to a new jar as an inoculation. Actual ended up being minimal splits every 3-4 weeks. Would add a second 20 drops of Micro Algae Grow if growth stalled. Color would be extremely dark green at time of split. Culture medium description: Current is 2l of ASW sterilized with chlorine at .8ml/2l , allowed to sit for 24hrs and then neutralized with equal parts sodium thiosulfate. 20 drops Micro Algae Grow added per 2l. Initial was 1 litre of artificial saltwater newly mixed and aged ~ 24hrs. Sterilized in microwave until the water boils in the jar. Allowed to cool at room temp. 10 drops (~1/2ml) Micro Algae Grow added to the culture.
